Soft ionization techniques like ESI and MALDI have enabled to study the structure and conformational dynamics of proteins using mass spectrometry platform. The present seminar describes the analysis of primary structure of hemoglobin variants in hemoglobinopathies and changes in quaternary structure of hemoglobin during its oxygenation across the globin chain peptide backbone. Using isotope exchange based mass spectrometry the conformational analysis of hemoglobin has been monitored inside the live Red Blood Cells. Structural analysis of hemoglobin has been found to be useful in the correct assessment of glycated hemoglobin, a gold standard biomarker for the diagnosis of diabetes mellitus. |
