Probing the structure of a membrane protein is a major challenge. One can exploit short range plasmonic fields around a nanoparticle, which can modulate fluorescence or Raman processes, to probe protein structures. However, till now it was not obvious how such plasmonic particles can probe membrane proteins. Here we show that lipid encased nanoparticles can enable such measurements on proteins attached to the lipid shell. Surface Enhanced Raman Spectroscopy can potentially determine the secondary structure, and fluorescence quenching can measure the relative depths of membrane insertion of different parts of a membrane protein. We employ this technique to discover that membrane inserted Amyloid-β oligomers have an anti-parallel beta sheet conformation and a preferred molecular orientation. |
